- Dimerization Domain (DD): E2Fs typically form heterodimers with DP proteins, enhancing their DNA-binding affinity and specificity.
- Transactivation Domain (TAD): Present in activator E2Fs (E2F1, E2F2, and E2F3a), this domain is responsible for recruiting co-activators and initiating transcription.
- RB-Binding Domain: E2F1 to E2F5 contain an RB-binding domain, allowing them to interact with retinoblastoma protein (RB) and related pocket proteins, which regulate E2F activity.
- Regulatory Domains: These regions are involved in post-translational modifications, such as phosphorylation, which modulate E2F activity and stability.
Classification and Subtypes
E2F transcription factors are classified into two main groups based on their function:
- Activator E2Fs: E2F1, E2F2, and E2F3a, which promote the transcription of genes required for S-phase entry and DNA replication.
- Repressor E2Fs: E2F3b, E2F4, E2F5, E2F6, E2F7, and E2F8, which primarily function to repress transcription and are involved in maintaining cell cycle arrest.

E2F transcription factors are involved in a wide array of cellular processes:
- Cell Cycle Regulation: Activator E2Fs induce the expression of genes necessary for DNA synthesis and cell cycle progression, while repressor E2Fs maintain quiescence by inhibiting these genes.
- Apoptosis: E2F1, in particular, can induce apoptosis by upregulating pro-apoptotic genes such as p53 and p73.
- DNA Damage Response: E2Fs are involved in the cellular response to DNA damage, ensuring that damaged DNA is repaired before cell division.
- Differentiation: E2Fs also play roles in cellular differentiation, with specific E2F family members being important in the differentiation of various cell types, including muscle and neuronal cells.
Clinical Issues
- Cancer: Dysregulation of E2F activity is a common feature in many cancers. Overexpression of activator E2Fs or loss of repressor E2Fs can lead to uncontrolled cell proliferation and tumor development. For example, E2F1 is often overexpressed in cancers such as breast, lung, and prostate cancers.
- Retinoblastoma: Mutations in the RB protein, which regulates E2F activity, are linked to the development of retinoblastoma, a rare childhood cancer of the retina.
- Therapeutic Targets: Given their central role in cell cycle regulation, E2Fs are considered potential targets for cancer therapy. Strategies to inhibit E2F activity or restore normal RB function are being explored in cancer treatment.
