Background
E2F7 is an atypical E2F transcription factor that functions as a repressor of E2F target genes, particularly those involved in DNA replication and cell cycle progression. Unlike classical E2Fs, E2F7 contains two DNA-binding domains but lacks a transactivation domain, enabling it to directly bind and repress target promoters without the involvement of RB proteins. E2F7 is upregulated during the G2/M phase and acts as a negative feedback regulator to suppress E2F-driven proliferation. It plays a key role in cellular stress responses, where it regulates genes involved in DNA damage repair, such as p53 target genes, to maintain genomic stability. Dysregulation of E2F7 contributes to tumorigenesis by disrupting cell cycle checkpoints and DNA damage responses. Knockout studies show increased sensitivity to DNA damage, defective cell cycle arrest, and aberrant proliferation, underscoring E2F7’s role in genomic maintenance, stress response, and tumor suppression.
