Background
E2F6 is a unique member of the E2F family that functions primarily as a transcriptional repressor independent of retinoblastoma (RB) proteins. Unlike other E2F family members, E2F6 lacks a transactivation domain and instead represses gene expression through association with polycomb group (PcG) proteins and chromatin-modifying complexes. E2F6 plays a critical role in gene silencing during the G1/S transition and is particularly involved in maintaining transcriptional repression during cell differentiation and development. It regulates genes involved in DNA repair, meiosis, and embryonic patterning by modulating chromatin structure. E2F6 is highly expressed in embryonic stem cells and germ cells, where it prevents aberrant transcription of proliferation-associated genes. Knockout studies reveal defects in meiosis and homeotic gene expression, highlighting its importance in chromatin remodeling, developmental regulation, and transcriptional repression.
