Background

E2F2 is another member of the E2F family that functions as a key transcriptional regulator of genes involved in cell cycle progression, particularly during the G1/S transition. Like E2F1, E2F2 is regulated by the retinoblastoma protein (pRB), which controls its ability to activate target genes necessary for DNA replication and mitosis. E2F2 plays a role in balancing cell proliferation and differentiation, particularly in hematopoietic and immune cells. Dysregulation of E2F2 contributes to hyperproliferation, genomic instability, and tumorigenesis, with studies showing its overexpression in cancers such as lymphoma and leukemia. Conversely, E2F2 can act as a tumor suppressor in specific contexts by inducing cell cycle arrest or apoptosis. Knockout models exhibit impaired T cell differentiation, deregulated cell growth, and altered immune homeostasis, highlighting E2F2’s role in cell cycle regulation, immune development, and cancer biology.
