Background

Contactins are a family of immunoglobulin-like cell adhesion molecules that play crucial roles in the nervous system, particularly in the development and maintenance of neuronal networks. Protein Structure

Contactins are glycosylphosphatidylinositol (GPI)-anchored proteins, meaning they are attached to the cell membrane via a GPI anchor rather than transmembrane domains. This anchorage allows them to interact with other cell surface molecules and the extracellular matrix. Structurally, contactins are characterized by:

  1. Immunoglobulin-like domains: Contactins typically have six immunoglobulin-like (Ig) domains at the N-terminal end. These domains are critical for protein-protein interactions and are involved in homophilic (same protein) and heterophilic (different protein) binding.
  2. Fibronectin type III domains: Following the Ig domains, contactins have four to five fibronectin type III (FNIII) domains. These domains contribute to the structural integrity and facilitate further interactions with other cellular components.
  3. GPI anchor: The C-terminal end of contactins is linked to the cell membrane via a GPI anchor. This unique feature allows contactins to be released from the membrane under certain conditions, providing flexibility in their function.


Classification and Subtypes

The contactin family can be classified based on their expression patterns and functional roles in the nervous system:

  • CNTN1 (Contactin-1): Involved in the formation of paranodal junctions in myelinated axons.
  • CNTN2 (Contactin-2/TAG-1): Plays a role in axonal growth and guidance.
