Descripción
Contactin 6 (CNTN6), also referred to as NB-3, is a neuronal cell adhesion molecule that is highly expressed in the brain and spinal cord during development. It regulates axon guidance, neuronal migration, and synapse formation, particularly in corticospinal tract development and cerebellar circuit organization. CNTN6 interacts with other contactins and neural receptors to facilitate synaptic connectivity and plasticity, contributing to the establishment of neural networks involved in motor control and coordination. Genetic studies have implicated CNTN6 in neurodevelopmental disorders such as autism spectrum disorders (ASD) and intellectual disability, where its dysregulation disrupts neuronal connectivity and cognitive functions. In addition to its roles in development, CNTN6 expression persists in the adult nervous system, suggesting ongoing functions in synaptic maintenance and plasticity. Structurally, CNTN6 features extracellular immunoglobulin-like and fibronectin type III domains, which mediate its adhesive and signaling interactions. Emerging research also highlights its potential involvement in neuropathological conditions, including neurodegenerative diseases and peripheral neuropathies.
