Family main features
Insulin Receptor Substrates (IRS) are a family of proteins that play a pivotal role in the insulin signaling pathway, crucial for regulating glucose homeostasis, metabolism, and cell growth. Upon insulin binding to its receptor (IR), the receptor undergoes autophosphorylation and subsequently phosphorylates IRS proteins on tyrosine residues. These phosphorylated tyrosine residues serve as docking sites for various SH2 domain-containing signaling molecules, thus propagating downstream signaling cascades such as the PI3K/Akt and Ras/MAPK pathways. There are several members of the IRS family, with IRS1 and IRS2 being the most studied due to their significant roles in metabolic regulation.

Insulin Receptor Substrates (IRS) are critical adaptor proteins in the insulin signaling pathway, essential for regulating glucose metabolism, growth, and survival. IRS proteins possess PH and PTB domains for membrane localization and interaction with the insulin receptor, along with multiple phosphorylation sites that serve as docking points for downstream signaling molecules. Dysregulation of IRS function is implicated in metabolic diseases such as type 2 diabetes and obesity, as well as in cancer and neurodegenerative diseases.
