  • Death Domain: A hallmark of many apoptotic proteins, the death domain in UNC5 receptors mediates apoptotic signaling pathways, particularly in the absence of netrin-1.
  • DCC-Interacting Domain: This domain interacts with DCC and other intracellular signaling molecules to modulate cellular responses to netrin-1.


Classification and Subtypes

The UNC5 family of receptors is classified into four main subtypes, each with unique and overlapping roles in development and disease:

  1. UNC5A: Predominantly expressed in the developing nervous system, UNC5A plays a role in axon guidance and neuronal migration.
  2. UNC5B: Widely expressed in the nervous system and vasculature, UNC5B is involved in both neuronal guidance and angiogenesis. It also plays a role in modulating immune cell function.

UNC5 receptors primarily mediate repulsive guidance cues in response to netrin-1. When netrin-1 binds to UNC5, it typically results in the repulsion of growth cones, steering developing axons away from regions expressing high levels of netrin-1. This mechanism ensures the proper wiring of neural circuits by preventing axons from entering inappropriate target areas.

In addition to their roles in axon guidance, UNC5 receptors are involved in:

  • Cell Migration: UNC5 receptors regulate the migration of various cell types, including neurons, endothelial cells, and immune cells. This function is crucial for tissue patterning and organogenesis.
  • Apoptosis: In the absence of netrin-1, UNC5 receptors can induce apoptosis through their intracellular death domains. This mechanism is thought to prevent cells from surviving in inappropriate locations, contributing to tissue homeostasis.
  • Angiogenesis: UNC5B, in particular, plays a significant role in vascular development by mediating the repulsion of endothelial cells during the formation of blood vessels.

Dysregulation of UNC5 receptors and their signaling pathways is implicated in various pathological conditions:

  • Cancer: Aberrant expression or mutation of UNC5 receptors has been linked to tumorigenesis. Loss of function or downregulation of UNC5 receptors can contribute to uncontrolled cell survival and migration, promoting cancer progression and metastasis. Conversely, overexpression of UNC5 receptors can induce apoptosis in cancer cells, suggesting their potential as therapeutic targets.
  • Neurodegenerative Diseases: Altered UNC5 signaling has been associated with neurodegenerative conditions such as Alzheimer's disease. Misregulation of axon guidance and neuronal survival pathways may contribute to the pathogenesis of these disorders.
