Description

Receptor for netrin required for axon guidance. Mediates axon repulsion of neuronal growth cones in the developing nervous system upon ligand binding. Axon repulsion in growth cones may be caused by its association with DCC that may trigger signaling for repulsion. Also involved in corticospinal tract axon guidances independently of DCC. It also acts as a dependence receptor required for apoptosis induction when not associated with netrin ligand (By similarity).

Descripción

unc-5 netrin receptor C (UNC5C) functions as a dependence receptor that mediates cellular responses to the presence or absence of netrin-1. UNC5C is critically involved in guiding axonal migration and positioning during neural development, playing a pivotal role in organizing the central and peripheral nervous systems. It primarily facilitates repulsive guidance signals, ensuring neurons and axons navigate away from inappropriate regions, thus contributing to the precision of neural circuit formation. Apart from its neural guidance role, UNC5C is implicated in maintaining tissue integrity by regulating cell adhesion and polarity. Its tumor suppressor function is mediated by the induction of apoptosis when netrin-1 is absent, which prevents uncontrolled cellular proliferation and migration. This receptor’s expression and function are often disrupted in cancers, where loss of UNC5C is linked to enhanced tumor invasiveness and resistance to programmed cell death. Furthermore, UNC5C plays roles in vascular development, ensuring proper endothelial cell organization and angiogenesis. Dysfunction of UNC5C signaling is also associated with neurodegenerative conditions, making it a subject of interest for therapeutic interventions targeting neural and oncological disorders.
