Description

UNC5D is receptor for netrin. May be involved in axon guidance by mediating axon repulsion of neuronal growth cones in the developing nervous system upon ligand binding. Axon repulsion in growth cones may be caused by its association with DCC that may trigger signaling for repulsion. It also acts as a dependence receptor required for apoptosis induction when not associated with netrin ligand (By similarity).

unc-5 netrin receptor D (UNC5D) operates as a dependence receptor for netrin-1, significantly contributing to axonal guidance, vascular organization, and cellular homeostasis. In the nervous system, UNC5D directs the repulsive migration of axons and neuronal processes, facilitating the formation of precise neural circuits during embryogenesis. Its function extends beyond the nervous system, as it modulates cell migration, adhesion, and apoptosis in a variety of tissue types. As a tumor suppressor, UNC5D induces apoptosis in the absence of netrin-1, acting as a checkpoint to inhibit malignant cell survival and growth. Dysregulation of UNC5D expression or signaling has been implicated in various cancers, including colorectal and lung carcinomas, where its loss allows tumor cells to evade apoptosis and metastasize. Additionally, UNC5D is involved in vascular development, where it ensures the proper formation and organization of blood vessels by mediating endothelial cell migration and positioning.
