Background
unc-5 netrin receptor D (UNC5D) operates as a dependence receptor for netrin-1, significantly contributing to axonal guidance, vascular organization, and cellular homeostasis. In the nervous system, UNC5D directs the repulsive migration of axons and neuronal processes, facilitating the formation of precise neural circuits during embryogenesis. Its function extends beyond the nervous system, as it modulates cell migration, adhesion, and apoptosis in a variety of tissue types. As a tumor suppressor, UNC5D induces apoptosis in the absence of netrin-1, acting as a checkpoint to inhibit malignant cell survival and growth. Dysregulation of UNC5D expression or signaling has been implicated in various cancers, including colorectal and lung carcinomas, where its loss allows tumor cells to evade apoptosis and metastasize.
