Background:

Opioid receptors are a family of G protein-coupled receptors (GPCRs) that mediate the effects of endogenous opioid peptides and exogenous opioid drugs. They play crucial roles in pain modulation, reward processing, and various physiological functions.

Opioid receptors bind to endogenous opioid peptides, including β-endorphin, enkephalins, dynorphins, and nociceptin/orphanin FQ. 1.   OPRD1 (Opioid Receptor Delta 1):

  • Physiological Functions: Mediates analgesia, modulation of emotional responses, and regulation of gastrointestinal motility.


2.   OPRK1 (Opioid Receptor Kappa 1):

  • Alias: Also known as κ-opioid receptor.
  • Background: OPRK1 is widely expressed in the brain, spinal cord, and peripheral tissues.
  • Physiological Functions: Involved in analgesia, stress responses, mood regulation, and dysphoria.


3.   OPRM1 (Opioid Receptor Mu 1):

  • Alias: Also known as μ-opioid receptor.
  • Background: OPRM1 is predominantly expressed in the central nervous system, including regions such as the brainstem, thalamus, and spinal cord.
  • Physiological Functions: Mediates analgesia, euphoria, respiratory depression, and reward reinforcement.


4.   OPRL1 (Opioid Receptor-Like 1):

  • Alias: Also known as nociceptin receptor or orphanin FQ receptor.
  • Background: OPRL1 is widely distributed in the brain, spinal cord, and peripheral tissues.
  • Physiological Functions: Binds to the endogenous neuropeptide nociceptin/orphanin FQ (N/OFQ) and modulates pain perception, stress responses, and mood regulation.
