Background

The ABCD subfamily is part of the ATP-binding cassette (ABC) transporter superfamily, which uses ATP hydrolysis to transport various molecules across membranes. ABCD transporters are primarily located in peroxisomes, where they play critical roles in the transport of fatty acids and other substrates necessary for peroxisomal metabolism.These transporters play a crucial role in the oxidation of a diverse range of metabolites, including saturated, monounsaturated, and polyunsaturated fatty acids, branched-chain fatty acids, bile acids, and dicarboxylic acids. Despite the impermeable barrier of the peroxisomal membrane to these metabolites, the mammalian peroxisomal membrane houses three ATP-binding cassette (ABC) half-transporters: ABCD1, ABCD2, and ABCD3. Functioning predominantly as homodimers, the ABCD transporters facilitate the transport of various acyl-CoAs


Protein Structure

ABCD transporters share common structural features typical of ABC proteins:

  • Transmembrane Domains (TMDs): ABCD proteins usually have six transmembrane helices that span the peroxisomal membrane and form the pathway for substrate transport.
  • Nucleotide Binding Domains (NBDs): These domains bind and hydrolyze ATP to drive the transport process. Each ABCD protein typically has one NBD, and they function as homodimers or heterodimers.


Classification and Subtypes

The ABCD family includes several key members, with notable examples being:

  • ABCD1: Also known as the adrenoleukodystrophy protein (ALDP), involved in the transport of very-long-chain fatty acids (VLCFAs) into peroxisomes.
  • ABCD2: Similar in function to ABCD1, but with a broader substrate specificity.
  • ABCD3: Also known as PMP70, involved in the transport of branched-chain fatty acids and other substrates into peroxisomes.
  • ABCD4: Plays a role in the transport of cobalamin (vitamin B12) into the peroxisome for processing.
