  • DNA-binding Domain (DBD): Comprises two zinc finger motifs that enable the receptor to bind to specific DNA sequences known as LXR response elements (LXREs) in the promoters of target genes.
  • Hinge Region: Connects the DBD to the ligand-binding domain and contributes to the receptor's structural flexibility and dimerization.
  • Ligand-binding Domain (LBD): Contains the activation function-2 (AF-2) region and is responsible for binding ligands (oxysterols). The LBD also facilitates the interaction with coactivators and corepressors to regulate gene transcription.


Classification and Subtypes

The LXR family includes:

  1. LXRα (NR1H3): Predominantly expressed in the liver, intestine, adipose tissue, and macrophages. It plays a critical role in regulating cholesterol, fatty acid, and glucose metabolism.
