Background

The Retinoid X Receptor (RXR) is a type of nuclear receptor that plays a central role in the regulation of gene expression involved in various physiological processes. RXRs are part of the nuclear receptor superfamily, which includes receptors for steroid hormones, thyroid hormone, retinoic acid, and other signaling molecules. RXRs serve as obligatory heterodimerization partners for several other nuclear receptors, enhancing their DNA-binding and transcriptional activities. These heterodimers include receptors such as peroxisome proliferator-activated receptors (PPARs), liver X receptors (LXRs), and vitamin D receptors (VDRs), among others. RXR's ability to form heterodimers makes it a pivotal node in the signaling pathways governing metabolism, cell differentiation, and homeostasis.


Protein Structure

RXRs consist of several functional domains:

  • DNA-Binding Domain (DBD): Contains two zinc finger motifs, enabling RXRs to bind to specific DNA sequences known as response elements.
  • Ligand-Binding Domain (LBD): This domain binds to retinoids, specifically 9-cis retinoic acid, and plays a crucial role in receptor activation and interaction with coactivators or corepressors.
  • Dimerization Domain: Facilitates the formation of homodimers or heterodimers with other nuclear receptors.
  • Activation Function (AF) Domains: The AF-1 domain is located at the N-terminus and is involved in ligand-independent transcriptional activation, while the AF-2 domain at the C-terminus mediates ligand-dependent activation.


Classification and Subtypes

RXRs are classified into three subtypes, each encoded by a distinct gene:

  • RXRα (NR2B1): Predominantly expressed in the liver, kidney, spleen, and other tissues.
  • RXRβ (NR2B2): Ubiquitously expressed across various tissues.
  • RXRγ (NR2B3): Primarily found in muscle and brain tissues.

  • Gene Regulation: RXRs regulate genes involved in diverse biological processes, including metabolism, cell proliferation, apoptosis, and differentiation.
