Nucleotide Binding Oligomerization Domain (NOD)-containing proteins are a class of intracellular pattern recognition receptors (PRRs) that play a critical role in the innate immune system.   • C-terminal Leucine-rich repeats (LRRs): These are involved in ligand recognition and binding. The LRRs are responsible for sensing specific microbial or host-derived molecules.
  • N-terminal Effector-binding domain: This domain can be a CARD (Caspase Activation and Recruitment Domain) or other similar motifs that mediate downstream signaling by interacting with other proteins involved in immune responses.

NOD-containing proteins are classified based on their structural and functional characteristics:

  • NOD1 and NOD2: These are the most well-characterized members of the NOD-like receptor (NLR) family. NOD1 recognizes γ-D-glutamyl-meso-diaminopimelic acid (a component of Gram-negative bacterial peptidoglycan), while NOD2 detects muramyl dipeptide (MDP), a component found in both Gram-positive and Gram-negative bacteria.

  • Pathogen Detection: They recognize intracellular PAMPs, leading to the activation of NF-κB and MAPK signaling pathways, which results in the production of pro-inflammatory cytokines and antimicrobial peptides.
  • Inflammatory Response: NOD proteins like NLRP3 are involved in the formation of inflammasomes, which activate inflammatory caspases (e.g., caspase-1) and promote the maturation of IL-1β and IL-18, key cytokines in inflammation.

  • Inflammatory Bowel Disease (IBD): Mutations in NOD2 are strongly associated with Crohn's disease, a type of IBD. These mutations can lead to inappropriate immune responses to gut microbiota.
  • Autoimmune Diseases: Dysregulation of NOD signaling pathways can contribute to autoimmune conditions, where the immune system erroneously attacks the body's own cells.
  • Infectious Diseases: Pathogens can evade detection or manipulate NOD signaling to promote their survival and replication within host cells.
  • Cancer: Chronic inflammation driven by aberrant NOD signaling can lead to an increased risk of cancer development, particularly in tissues frequently exposed to pathogens or other inflammatory stimuli.
