Background

Fibroblast Growth Factor Receptor Substrates (FRS) are adaptor proteins that play critical roles in signal transduction pathways mediated by fibroblast growth factor receptors (FGFRs). FRS proteins share several conserved structural features that enable their function as adaptor proteins:

  • Phosphotyrosine-Binding (PTB) Domain: Located at the N-terminus, the PTB domain binds to specific phosphotyrosine motifs on activated FGFRs and other receptor tyrosine kinases, anchoring FRS proteins to the receptors.
  • Multiple Tyrosine Phosphorylation Sites: The C-terminal region of FRS proteins contains several tyrosine residues that, when phosphorylated by FGFRs, serve as docking sites for SH2 domain-containing signaling proteins, such as Grb2 and Shp2.
  • Myristoylation Sites: These sites facilitate membrane localization, which is crucial for the interaction of FRS proteins with membrane-bound receptors and signaling complexes.


Classification and Subtypes

The FRS family consists of two main proteins:

  • FRS2 (FRS2α): Ubiquitously expressed and involved in mediating FGFR signaling in various cell types, playing a key role in developmental processes and cellular responses to growth factors.
  • FRS3 (FRS2β): Structurally similar to FRS2 but with a more limited expression pattern and potentially distinct roles in specific tissues and cellular contexts.

FRS proteins are essential mediators of FGFR signaling and are involved in various biological processes:
