Allograft Inflammatory Factor 1 (AIF1), also known as Iba1 (ionized calcium-binding adapter molecule 1), is a protein primarily expressed in macrophages and microglia, the immune cells of the central nervous system. AIF1 is involved in various immune-related processes, including inflammation, antigen presentation, phagocytosis, and tissue repair. It plays a crucial role in the activation and migration of macrophages and microglia in response to injury, infection, or inflammation. AIF1 is highly expressed in activated macrophages and microglia, where it serves as a marker for these cells. It is also expressed in other cell types, including dendritic cells, certain subsets of T cells, and fibroblasts, albeit at lower levels. AIF1 has been implicated in various inflammatory and autoimmune diseases. Elevated levels of AIF1 expression are often observed in tissues affected by inflammation, such as in rheumatoid arthritis, multiple sclerosis, and atherosclerosis. In neuroinflammatory conditions, AIF1 expression is upregulated in microglia, contributing to the inflammatory response and neuronal damage. Regulation of Microglial Activation: In the central nervous system, AIF1 is particularly important for regulating the activation state of microglia, the resident immune cells. Upon activation, microglia upregulate AIF1 expression, which is associated with morphological changes and increased phagocytic activity. Dysregulated AIF1 expression in microglia has been linked to neuroinflammation and neurodegenerative diseases.
