Background

Acetyl Coenzyme A Carboxylase (ACAC) is a crucial enzyme in fatty acid metabolism, playing a key role in the regulation of fatty acid synthesis. It catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the first committed step in fatty acid biosynthesis. ACACA (Alpha), also known as ACC1, is primarily found in the cytoplasm. Unlike ACACA, which generates malonyl-CoA in the cytoplasm for fatty acid synthesis, ACACB produces malonyl-CoA in mitochondria, where it acts as an allosteric inhibitor of carnitine palmitoyltransferase 1 (CPT1). CPT1 is a key enzyme involved in the transport of fatty acids into the mitochondria for oxidation. By inhibiting CPT1, ACACB helps to regulate fatty acid oxidation based on cellular energy needs. Both isoforms of Acetyl-CoA Carboxylase are subject to complex regulation at multiple levels, including transcriptional, post-translational, and allosteric regulation, to ensure that fatty acid synthesis and oxidation are coordinated according to cellular energy demands and nutritional status.

  • ACACB (Beta): Mainly expressed in oxidative tissues such as muscle and heart. It regulates the rate of fatty acid oxidation by controlling the levels of malonyl-CoA, an inhibitor of carnitine palmitoyltransferase 1 (CPT1), which is crucial for fatty acid transport into mitochondria for oxidation.
