Background:

Leukotriene receptors are G protein-coupled receptors (GPCRs) that respond to leukotrienes, which are lipid mediators derived from arachidonic acid metabolism. LLTB4R-1 (Leukotriene B4 Receptor 1):

LTB4R-1 is a high-affinity receptor for leukotriene B4 (LTB4), a potent chemoattractant and pro-inflammatory mediator. LTB4R-1 is expressed in immune cells such as neutrophils, macrophages, and dendritic cells, as well as in other tissues including the skin and gastrointestinal tract.


Physiological Functions:

  • Mediates neutrophil chemotaxis and activation, contributing to inflammation and host defense responses.
  • Plays roles in allergic reactions, tissue repair, and the pathogenesis of inflammatory diseases such as asthma, inflammatory bowel disease (IBD), and psoriasis.

 

Clinical Implications:

Targeting LTB4R-1 signaling is a potential therapeutic strategy for inflammatory diseases and disorders characterized by excessive leukotriene production and neutrophil infiltration.


Cysteinyl Leukotriene Receptors:


CYSLTR1 (Cysteinyl Leukotriene Receptor 1):

Is a receptor for cysteinyl leukotrienes (CysLTs), including LTC4, LTD4, and LTE4, which are involved in allergic and inflammatory responses. CYSLTR1 is expressed in various tissues, including smooth muscle cells, endothelial cells, and immune cells such as mast cells and eosinophils.

      

Physiological Functions:

  • Mediates bronchoconstriction, vascular permeability, mucus secretion, and inflammatory responses in the respiratory tract.
  • Contributes to the pathogenesis of asthma, allergic rhinitis, and other allergic and inflammatory diseases.


Clinical Implications:

CYSLTR1 antagonists (e.g., montelukast) are used clinically as anti-asthmatic drugs and for the treatment of allergic rhinitis and exercise-induced bronchoconstriction.


CYSLTR2 (Cysteinyl Leukotriene Receptor 2):

Is another receptor for cysteinyl leukotrienes, sharing structural and functional similarities with CYSLTR1. CYSLTR2 is also expressed in various tissues and cell types, including smooth muscle cells, endothelial cells, and immune cells.

   

Physiological Functions:

  • Mediates similar responses to CYSLTR1, including bronchoconstriction, vascular permeability, and inflammatory processes.
  • May have overlapping or distinct roles compared to CYSLTR1 in specific tissues or cell types.

OXE Receptor:

OXER1 is a receptor for 5-oxo-eicosatetraenoic acid (5-oxo-ETE), a metabolite of arachidonic acid produced by 5-lipoxygenase. OXER1 is expressed in various tissues, including immune cells, endothelial cells, and smooth muscle cells.


Physiological Functions:

  • Regulates immune cell chemotaxis, inflammation, and vascular permeability in response to 5-oxo-ETE.
  • May play roles in allergic reactions, asthma, and cardiovascular diseases.


Clinical Implications:

OXER1 antagonists are being investigated for their potential therapeutic applications in inflammatory diseases and disorders associated with 5-oxo-ETE signaling.
