Background

Induced myeloid leukemia cell differentiation protein Mcl-1 is a member of the Bcl-2 family of proteins, which play a crucial role in the regulation of apoptosis. Mcl-1 was initially identified due to its upregulation during the differentiation of myeloid leukemia cells. It is encoded by the MCL1 gene, which is located on chromosome 1q21. This protein is highly expressed in various tissues, including the heart, lungs, and liver, and is involved in promoting cell survival by inhibiting apoptosis. Mcl-1 is an essential protein for normal development, tissue homeostasis, and the immune response.


Protein Structure

  • Molecular Weight: Approximately 37 kDa.
  • Domains: Mcl-1 contains Bcl-2 homology (BH) domains, including BH1, BH2, BH3, and BH4. These domains facilitate its interactions with other Bcl-2 family members.
  • Structure: Mcl-1 is an anti-apoptotic protein with an N-terminal transmembrane domain that anchors it to the outer mitochondrial membrane, where it interacts with pro-apoptotic proteins to inhibit apoptosis.
  • Isoforms: Alternative splicing of the MCL1 gene produces multiple isoforms with distinct functions, including an anti-apoptotic long form (Mcl-1L) and a pro-apoptotic short form (Mcl-1S).

Mcl-1 is classified as an anti-apoptotic member of the Bcl-2 family. This family is divided into three main groups based on function:

  2. Pro-apoptotic effector proteins: Such as Bax and Bak.

  • Anti-Apoptotic Function: Mcl-1 inhibits apoptosis by sequestering pro-apoptotic proteins such as Bax and Bak, preventing them from inducing mitochondrial outer membrane permeabilization (MOMP) and subsequent cytochrome c release.
  • Cell Survival: Mcl-1 is critical for the survival of various cell types, including hematopoietic stem cells, cardiomyocytes, and neurons. It supports cell viability under conditions of stress, such as nutrient deprivation and DNA damage.
  • Immune Response: Mcl-1 is important for the development and function of immune cells, including neutrophils, T cells, and B cells. It helps maintain the balance between cell survival and cell death during immune responses.


Clinical Issues

  • Cancer: Overexpression of Mcl-1 is commonly observed in various cancers, including leukemia, lymphoma, and solid tumors. This overexpression contributes to cancer cell survival, resistance to chemotherapy, and poor prognosis. Mcl-1 inhibitors are being developed as potential cancer therapeutics.
  • Cardiovascular Diseases: Mcl-1 is essential for cardiac myocyte survival, and its dysregulation can contribute to heart diseases. Loss of Mcl-1 function in cardiomyocytes can lead to increased apoptosis and heart failure.
  • Neurodegenerative Diseases: Mcl-1 plays a role in protecting neurons from apoptosis. Its dysregulation is implicated in neurodegenerative conditions such as Alzheimer's disease.
