Background

Extracellular Domain (ECD):

  • The extracellular domain, which is approximately 180 amino acids in length, is responsible for ligand binding. This domain contains four cysteine-rich domains (CRDs), which are crucial for binding TNF-α with high affinity.
  • These CRDs maintain the structure of the extracellular region and mediate trimerization, which is essential for initiating signaling pathways.
  • The ligand-binding capability of the extracellular domain is facilitated by disulfide bridges within these cysteine-rich domains, ensuring structural integrity and correct receptor conformation for efficient TNF-α binding.

Transmembrane Domain (TMD):

Intracellular Domain (ICD):

  • Overactivation of TNFR1 is associated with chronic inflammatory and autoimmune diseases, such as rheumatoid arthritis, inflammatory bowel disease (IBD), systemic lupus erythematosus (SLE), and psoriasis. The excessive TNFR1-mediated NF-κB activation leads to sustained inflammation and tissue damage.
