Protein Structure

CD40LG is a type II transmembrane protein composed of approximately 261 amino acids with a molecular weight of about 33 kDa. It features three primary domains:

Extracellular Domain:

  • The extracellular region of CD40LG, approximately 215 amino acids in length, is responsible for binding to CD40 and initiating downstream signaling pathways. This region includes a TNF homology domain (THD), which adopts a trimeric beta-sandwich fold typical of TNF superfamily members.
  • The TNF homology domain is critical for receptor binding and is composed of antiparallel beta strands arranged in a compact structure, allowing for the formation of stable trimeric structures on the cell surface.
  • CD40LG is initially expressed as a transmembrane protein but can also be cleaved into a soluble form, which retains the ability to interact with CD40, though with potentially altered functional effects.

Transmembrane Domain:

  • CD40LG contains a single-pass transmembrane helix that anchors it in the plasma membrane, allowing it to function as a membrane-bound cytokine.
  • This domain stabilizes the protein within the membrane, facilitating receptor-ligand interactions at the cell surface and allowing CD40LG to mediate cell-cell interactions efficiently.

Cytoplasmic Domain:

  • The cytoplasmic domain is relatively short and plays a minimal role in signaling, as CD40LG primarily functions by binding to CD40 rather than signaling independently.
  • However, the cytoplasmic tail is necessary for the proper cellular localization and surface expression of CD40LG, and it plays a role in intracellular trafficking.


Classification and Subtypes

CD40LG is classified as a member of the tumor necrosis factor (TNF) ligand superfamily, specifically as TNFSF5. It does not have distinct subtypes but can exist in membrane-bound and soluble forms. The TNF superfamily also includes ligands such as TNF-α, CD27L, and CD30L, which are structurally and functionally related but distinct in their specific receptor interactions and downstream effects.

The CD40LG-CD40 axis is implicated in a range of clinical conditions due to its extensive involvement in immune regulation:

X-Linked Hyper-IgM Syndrome (XHIM):

  • Mutations in the CD40LG gene lead to X-linked hyper-IgM syndrome, a primary immunodeficiency disorder characterized by elevated levels of IgM and reduced levels of other immunoglobulins (IgG, IgA, IgE). Individuals with XHIM are susceptible to recurrent infections due to impaired B-cell function and a lack of immunoglobulin class-switching.
  • Patients with XHIM require lifelong immunoglobulin replacement therapy to manage infections and improve immune function.

Autoimmune Diseases:
