AGE - Advanced Glycation End Product (Receptor) | Elisa - Clia - Antibody - Protein
Family main features
Background:
Advanced glycation end products (AGEs) are a complex group of compounds that form through non-enzymatic reactions between reducing sugars and proteins, lipids, or nucleic acids. These reactions, known as glycation or Maillard reactions, occur spontaneously and accelerate under conditions of hyperglycemia (elevated blood sugar levels) and oxidative stress. AGEs accumulate in various tissues over time, particularly in long-lived proteins like collagen, and their formation is associated with aging and several chronic diseases, including diabetes, cardiovascular disease, Alzheimer's disease, and kidney disease. AGEs are formed through a series of chemical reactions between reducing sugars and proteins, lipids, or nucleic acids. These reactions result in the irreversible modification of biomolecules, leading to changes in their structure and function. AGEs accumulate in tissues over time, particularly in long-lived proteins such as collagen. This accumulation contributes to the pathogenesis of various age-related diseases. AGEs can cross-link with proteins, leading to the formation of advanced glycation end product-crosslinks (AGE-crosslinks). This cross-linking alters the structure and function of proteins and contributes to tissue stiffness and dysfunction. AGEs can also bind to cell surface receptors known as receptor for advanced glycation end products (RAGEs), activating signaling pathways that promote inflammation, oxidative stress, and tissue damage. AGEs can induce oxidative stress by generating reactive oxygen species (ROS) and impairing antioxidant defenses, leading to cellular damage and dysfunction. AGE accumulation and RAGE activation have been implicated in the pathogenesis of various diseases, including diabetes complications (e.g., nephropathy, retinopathy), cardiovascular disease (e.g., atherosclerosis), neurodegenerative diseases (e.g., Alzheimer's disease), and chronic kidney disease.
Protein Structure:
AGEs are diverse molecules resulting from the reaction between reducing sugars and the amino groups of proteins, lipids, or nucleic acids. Common structural features include:
- Cross-Linking: AGEs often form cross-links between molecules, altering their structure and function.
- Fluorescent Properties: Many AGEs exhibit fluorescence due to their complex molecular structures.
- Heterogeneous Composition: AGEs comprise a wide variety of molecular species, such as carboxymethyl-lysine (CML), pentosidine, and pyrraline.
Classification and Subtypes:
AGEs can be classified based on their origin and chemical properties:
- Endogenous AGEs: Formed naturally within the body through metabolic processes.
- Exogenous AGEs: Derived from external sources such as diet (e.g., cooked foods) and tobacco smoke.
- Types of AGEs: Specific types include CML, methylglyoxal-derived hydroimidazolones (MG-H1), and glucosepane.
Function and Biological Significance:
AGEs have significant biological effects due to their ability to modify proteins and interact with cellular receptors:
- Protein Modification: AGEs alter the structure and function of proteins, impairing their normal biological activities.
- Receptor Interaction: AGEs bind to receptors such as RAGE (Receptor for Advanced Glycation End Products), triggering inflammatory responses and oxidative stress.
- Tissue Damage: Accumulation of AGEs contributes to tissue stiffness, reduced elasticity, and impaired function, particularly in the cardiovascular system, kidneys, and eyes.
Interactions:
AGEs interact with various cellular components and influence multiple pathways:
- RAGE: Binding to RAGE activates signaling pathways that lead to inflammation, oxidative stress, and cellular dysfunction.
- Scavenger Receptors: Other receptors involved in the clearance of AGEs include macrophage scavenger receptors.
- Proteins and Enzymes: AGEs modify proteins and enzymes, disrupting their normal functions and contributing to disease pathogenesis.
Clinical Issues:
The accumulation of AGEs is associated with various chronic diseases and aging:
- Diabetes: Elevated blood sugar levels in diabetes accelerate AGE formation, contributing to complications such as neuropathy, retinopathy, and nephropathy.
- Cardiovascular Diseases: AGEs contribute to atherosclerosis, hypertension, and heart failure by promoting inflammation and vascular stiffness.
- Neurodegenerative Disorders: AGEs are implicated in the development and progression of Alzheimer's disease and other neurodegenerative conditions.
Summary:
Advanced Glycation End Products (AGEs) are harmful compounds formed through the non-enzymatic glycation of proteins, lipids, and nucleic acids. AGEs accumulate in tissues and contribute to aging and the pathogenesis of various chronic diseases, including diabetes, cardiovascular diseases, and neurodegenerative disorders. AGEs exert their effects by modifying proteins and interacting with cellular receptors such as RAGE, leading to inflammation, oxidative stress, and tissue damage. Understanding the role of AGEs in disease can inform strategies for prevention and treatment, highlighting the importance of managing blood sugar levels and reducing dietary intake of AGEs.
