SERPINB5, also known as maspin (mammary serine protease inhibitor), is a member of the serpin (serine protease inhibitor) superfamily. Unlike many other serpins that function as protease inhibitors, SERPINB5 is unique in that it does not inhibit protease activity. Discovered in the early 1990s, SERPINB5 has since been the subject of extensive research due to its involvement in various cellular processes and its implications in cancer biology. This protein is encoded by the SERPINB5 gene located on chromosome 18q21.3, a region that has been associated with several types of cancer, particularly breast cancer. SERPINB5 is primarily expressed in epithelial cells and has been implicated in processes such as cell adhesion, migration, and apoptosis.
SERPINB5 is a non-inhibitory serpin, a class of proteins that typically inhibit serine proteases through a suicide substrate mechanism. However, SERPINB5 lacks the critical reactive center loop (RCL) that is necessary for this inhibitory activity, setting it apart from classical serpins. The protein is approximately 42 kDa in size and comprises 375 amino acids.
