Descripción

PLEKHA5 is a lipid-binding adaptor protein that contains a pleckstrin homology (PH) domain, allowing it to interact with specific phosphoinositides, such as PI(3,4)P2 and PI(4,5)P2, at cellular membranes. It plays a role in intracellular trafficking, cell migration, and signal transduction, linking phosphoinositide dynamics to cytoskeletal reorganization and membrane recruitment of signaling proteins. PLEKHA5 is highly expressed in brain and neuronal tissues, where it participates in processes like synaptic signaling, axon guidance, and neuronal migration. Dysregulation of PLEKHA5 is associated with metastatic melanoma, where it enhances tumor cell invasion and migration through actin remodeling and signaling pathways like PI3K/Akt. Functional studies show that depletion of PLEKHA5 reduces metastatic potential and disrupts membrane-associated signaling, underscoring its role in cell polarity, phosphoinositide signaling, and cancer progression.
