Descripción
PLEKHA3 is a member of the pleckstrin homology (PH) domain-containing protein family that binds phosphoinositides, including PI(3,4)P2 and PI(4,5)P2, to regulate membrane targeting and intracellular signaling pathways. It is involved in processes such as cell polarity, membrane trafficking, and cytoskeletal organization, where it recruits signaling complexes to membrane compartments. PLEKHA3 is expressed in various tissues, including immune and epithelial cells, where it regulates endocytic vesicle transport, signal transduction, and cellular adhesion. It has been implicated in processes like epithelial-mesenchymal transition (EMT) and cancer progression, where its dysregulation promotes cell migration, invasion, and altered signaling dynamics. Knockdown studies reveal defects in endocytosis, impaired membrane recruitment of signaling proteins, and disrupted cytoskeletal organization, underscoring its critical role in phosphoinositide-mediated cellular signaling, vesicular trafficking, and cell polarity.
