Background
The CD2 molecule, also known as T-cell surface antigen T11/Leu-5, is a cell adhesion molecule found on the surface of T cells, natural killer (NK) cells, and thymocytes. CD2 is part of the immunoglobulin (Ig) superfamily and plays a crucial role in the immune response by facilitating cell-cell adhesion and signal transduction. It was one of the first T-cell markers identified and has been extensively studied due to its importance in T cell and NK cell functions.
Protein Structure
The CD2 molecule is a type I transmembrane glycoprotein with a molecular weight of approximately 50 kDa. Its structure includes:
- Extracellular Domain: The extracellular region of CD2 contains two immunoglobulin-like domains—one variable (V) domain and one constant (C2) domain. This configuration is typical of members of the Ig superfamily and is crucial for the molecule’s role in cell adhesion and interaction with ligands.
- Transmembrane Domain: This hydrophobic domain anchors CD2 in the plasma membrane, allowing it to function as a receptor and signaling molecule.
- Intracellular Domain: The cytoplasmic tail of CD2 is involved in signal transduction. It contains several conserved motifs and phosphorylation sites that interact with intracellular signaling molecules, such as protein kinases and adaptor proteins.
The structure of CD2 enables it to mediate various interactions and signaling events that are essential for T cell activation and function.
Classification and Subtypes
CD2 is a member of the immunoglobulin superfamily and does not have subtypes like some other cell surface molecules. However, it interacts with multiple ligands and receptors, including:
- CD58 (LFA-3): A ligand found on antigen-presenting cells (APCs) and other cell types that bind to CD2 and facilitate T cell-APC adhesion.
- CD48: Another ligand for CD2, primarily found on hematopoietic cells, that plays a role in T cell activation and interaction with other immune cells.

CD2 plays several critical roles in the immune system:

The CD2 molecule has been implicated in various clinical conditions and therapeutic applications:
- Autoimmune Diseases: Dysregulation of CD2 expression or function can contribute to autoimmune diseases. For example, aberrant CD2 signaling has been associated with conditions such as multiple sclerosis and rheumatoid arthritis. Understanding CD2’s role in these diseases could lead to new therapeutic strategies.
