AMPH - Amphiphysin | Elisa - Clia - Antibody - Protein

Family main features

Background:

AMPH or Amphiphysin, is a protein that plays important roles in the nervous system, particularly in synaptic vesicle endocytosis and membrane trafficking. Amphiphysin is a cytosolic protein found in nerve terminals, especially in the brain. It belongs to a family of proteins that contain an N-terminal BAR (Bin-Amphiphysin-Rvs) domain, a central coiled-coil domain, and a C-terminal SH3 (Src homology 3) domain. One of the main functions of Amphiphysin is its involvement in synaptic vesicle endocytosis, a process crucial for maintaining neurotransmission at synapses. It helps in the formation of clathrin-coated vesicles, which are responsible for internalizing neurotransmitter-containing vesicles after they have released their contents into the synaptic cleft. The BAR domain of Amphiphysin allows it to sense and induce membrane curvature. This ability is important for its role in shaping membranes during vesicle formation and trafficking. Amphiphysin interacts with dynamin, another protein involved in vesicle formation and fission. This interaction is crucial for the scission of clathrin-coated vesicles from the plasma membrane during endocytosis. Apart from its role in vesicle trafficking, Amphiphysin is also involved in neuronal signaling and cytoskeletal regulation. It interacts with various proteins involved in actin dynamics and synaptic function. Autoantibodies against Amphiphysin have been found in patients with neurological disorders, such as stiff-person syndrome (SPS) and paraneoplastic neurological syndromes. These disorders involve abnormal muscle stiffness and other neurological symptoms.

Protein Structure:

  • BAR Domain: Amphiphysin contains a Bin-Amphiphysin-Rvs (BAR) domain, which is responsible for binding to curved membranes and inducing membrane curvature.
  • SH3 Domain: The Src homology 3 (SH3) domain facilitates interactions with proline-rich motifs found in various endocytic proteins, including dynamin.
  • Amphipathic Helices: These regions help in membrane association and curvature generation.

Classification and Subtypes:

  • Family: Amphiphysin belongs to the BAR domain-containing protein family, known for their roles in membrane dynamics and cytoskeleton interactions.
  • Isoforms: There are two main isoforms of amphiphysin:
  • Amphiphysin I (AMPH1): Primarily found in the brain and involved in synaptic vesicle endocytosis.
  • Amphiphysin II (BIN1): More ubiquitously expressed and has roles in muscle development, membrane trafficking, and possibly tumor suppression.

Function and Biological Significance:

  • Synaptic Vesicle Endocytosis: Amphiphysin I plays a crucial role in synaptic vesicle recycling by interacting with dynamin, clathrin, and other endocytic proteins, thus facilitating the retrieval of synaptic vesicles after neurotransmitter release.
  • Membrane Curvature and Dynamics: The BAR domain allows amphiphysin to sense and induce membrane curvature, which is vital for the formation of endocytic vesicles.
  • Muscle Development: Amphiphysin II is involved in T-tubule formation in muscle cells, which is essential for muscle contraction.

Interactions:

  • Dynamin: Amphiphysin binds to dynamin through its SH3 domain, promoting the scission of clathrin-coated vesicles.
  • Clathrin and AP2: These interactions help in recruiting amphiphysin to sites of endocytosis.
  • Other BAR Domain Proteins: Amphiphysin forms heterodimers with other BAR domain proteins to modulate membrane curvature.

Clinical Issues:

  • Autoimmune Disorders: Autoantibodies against amphiphysin are found in patients with paraneoplastic syndromes, particularly paraneoplastic stiff-person syndrome, often associated with breast cancer.
  • Neurological Disorders: Mutations or dysregulation in amphiphysin II (BIN1) are linked to centronuclear myopathy, a congenital muscle disorder, and have been implicated in Alzheimer's disease due to its role in membrane trafficking and potentially in tau pathology.
  • Cancer: Alterations in BIN1 expression have been observed in various cancers, suggesting a potential tumor suppressor role.

Summary:

Amphiphysin is a BAR domain-containing protein essential for synaptic vesicle endocytosis, membrane dynamics, and muscle development. It interacts with key endocytic proteins like dynamin and clathrin, facilitating vesicle formation and retrieval. Amphiphysin is involved in critical cellular processes and has significant implications in neurological disorders, autoimmune diseases, and cancer. Understanding its function and regulation offers insights into these pathologies and potential therapeutic targets.
