Descripción
TMED5 is a member of the p24 trafficking protein family that regulates protein transport and sorting between the ER and the Golgi apparatus. It functions as a cargo receptor by recognizing and packaging proteins into COPII-coated vesicles for export from the ER, while facilitating vesicle retrieval back to the ER through interactions with COPI machinery. TMED5 maintains ER-Golgi communication and contributes to protein quality control, ensuring proper folding and maturation of secretory and membrane proteins. It is expressed in multiple tissues and is essential for cellular homeostasis, especially in rapidly growing or secretory cells. TMED5 also plays a role in regulating growth factor signaling pathways and maintaining Golgi structure. Dysregulation of TMED5 has been linked to developmental defects and cancers due to altered trafficking dynamics and impaired protein secretion. Knockdown studies demonstrate defective vesicle transport, Golgi fragmentation, and disrupted secretory pathway balance, emphasizing its role in protein trafficking, organelle integrity, and cellular signaling.
