Descripción

SHC2, also known as SCK, is an adaptor protein primarily expressed in neural tissues, where it mediates signaling downstream of receptor tyrosine kinases and neurotrophin receptors, including TrkA/B. Like SHC1, it contains PTB and SH2 domains that facilitate interactions with phosphorylated receptors, activating Ras/MAPK and PI3K/Akt pathways to regulate processes such as neuronal survival, differentiation, and synaptic plasticity. SHC2 is essential for neurotrophin signaling, supporting neural development and maintenance of neuronal connections in the brain. Dysregulation of SHC2 has been associated with neurodevelopmental and neurodegenerative disorders, including impaired neuronal survival and axon guidance. Studies suggest that altered SHC2 expression may also contribute to certain cancers through its ability to regulate mitogenic and survival signaling pathways. Knockout models display defects in neuronal signaling, synapse formation, and brain development, highlighting its specialized role in the nervous system and neurotrophin-mediated signaling pathways.
