AGR2 is a protein involved in the endoplasmic reticulum (ER) stress response and functions as a protein disulfide isomerase (PDI), facilitating the proper folding of secreted and membrane proteins. It is primarily expressed in secretory tissues such as the gastrointestinal tract, lungs, and prostate, where it maintains ER homeostasis by catalyzing the formation of disulfide bonds in proteins. AGR2 also plays a role in cell adhesion and migration, partly through its interaction with mucins like MUC1 and MUC2. Overexpression of AGR2 is frequently observed in cancers, including breast, pancreatic, and prostate cancers, where it contributes to tumor progression, chemoresistance, and metastasis by promoting cell survival under stress conditions
