Background

AKAP6, also known as mAKAP (muscle AKAP), is a scaffold protein predominantly expressed in cardiac and skeletal muscle cells. It anchors PKA and other signaling molecules, such as phosphodiesterases (PDEs) and calcium-handling proteins, to the nuclear envelope, creating a signaling hub that regulates hypertrophic responses, calcium dynamics, and transcription. AKAP6 is critical for cardiomyocyte function, as it spatially coordinates cAMP signaling and calcium release to control excitation-contraction coupling and cardiac remodeling under stress. It interacts with PDE4D to regulate local cAMP levels and prevent hyperactivation of PKA, ensuring precise signaling.
