Descripción
AKAP6, also known as mAKAP (muscle AKAP), is a scaffold protein predominantly expressed in cardiac and skeletal muscle cells. It anchors PKA and other signaling molecules, such as phosphodiesterases (PDEs) and calcium-handling proteins, to the nuclear envelope, creating a signaling hub that regulates hypertrophic responses, calcium dynamics, and transcription. AKAP6 is critical for cardiomyocyte function, as it spatially coordinates cAMP signaling and calcium release to control excitation-contraction coupling and cardiac remodeling under stress. It interacts with PDE4D to regulate local cAMP levels and prevent hyperactivation of PKA, ensuring precise signaling. Mutations or dysfunctions in AKAP6 are associated with pathological cardiac hypertrophy and heart failure. Additionally, AKAP6 integrates signals from beta-adrenergic receptors, impacting gene transcription through interactions with nuclear factor NFAT and histone deacetylases. AKAP6’s unique role in organizing nuclear and perinuclear signaling makes it vital for cardiac development and adaptive responses to physiological stress.
