Prostaglandin F2-Alpha Receptor (PTGFR), also known as the FP receptor, is a G protein-coupled receptor activated by prostaglandin F2-alpha (PGF2α), a lipid-derived signaling molecule involved in reproductive, vascular, and inflammatory processes. PTGFR primarily couples to Gq proteins, activating phospholipase C (PLC), increasing intracellular calcium, and stimulating downstream signaling pathways, including Rho-kinase and MAPK. PTGFR is widely expressed in the uterus, corpus luteum, vascular smooth muscle, and ocular tissues, where it regulates smooth muscle contraction, luteolysis, and intraocular pressure. In the female reproductive system, PTGFR mediates uterine contraction during labor and luteolysis, the regression of the corpus luteum, which is essential for the estrous cycle. Clinically, FP receptor agonists such as latanoprost and bimatoprost are widely used to treat glaucoma by reducing intraocular pressure through enhanced aqueous humor outflow. Dysregulation of PTGFR signaling is associated with preterm labor, dysmenorrhea, and elevated intraocular pressure. Due to its role in smooth muscle regulation and ocular function, PTGFR remains a critical therapeutic target for reproductive and ophthalmic disorders.
