Descripción
PHF3 is a chromatin-binding protein containing a PHD domain that mediates the recognition of methylated histones, particularly H3K4me3, to regulate transcriptional activation and RNA polymerase II-dependent transcription. It serves as a transcriptional regulator by linking histone modifications to transcription elongation, ensuring precise gene expression during cellular proliferation and differentiation. PHF3 is expressed in neuronal and hematopoietic tissues, where it contributes to neurodevelopment, DNA damage repair, and cell survival. It is also involved in tumor suppression, where it stabilizes chromatin structure and controls the expression of growth-regulating genes. Dysregulation of PHF3 is linked to neurodevelopmental disorders and cancers, where its loss of function disrupts transcriptional regulation and genome stability. Knockout studies reveal defects in transcription elongation, impaired DNA repair, and disrupted cellular differentiation, underscoring its critical role in transcriptional fidelity, chromatin regulation, and cellular homeostasis.
