The Neuromedin B receptor (NMBR) is a G-protein-coupled receptor (GPCR) that binds to neuromedin B (NMB), a peptide belonging to the bombesin-like peptide family. NMBR is widely expressed in the central nervous system (CNS), the gastrointestinal tract, and the lungs. In the CNS, NMBR plays a role in thermoregulation, particularly in body temperature control, by modulating hypothalamic pathways. It is also implicated in feeding behavior and satiety regulation, as it interacts with neuroendocrine circuits that suppress appetite. Beyond the CNS, NMBR influences smooth muscle contraction in the gastrointestinal tract, supporting motility and digestion processes. Its expression in the lungs contributes to pulmonary regulation, including bronchial smooth muscle contraction. Dysregulation of NMBR signaling has been linked to certain neurological disorders and cancer progression, especially in lung and prostate cancers, where it stimulates cell proliferation and migration. NMBR also modulates hormone secretion, including pituitary hormone release, further linking it to neuroendocrine functions. The receptor is currently being explored as a potential therapeutic target for disorders related to appetite regulation, obesity, and specific cancers, given its involvement in growth signaling pathways and cell division.
