Description

Once phosphorylated, functions as an adapter protein in signal transduction cascades by binding to SH2 and SH3 domain-containing proteins. Role in G2-M progression in the cell cycle. Represses CBP-dependent transcriptional activation apparently by competing with other nuclear factors for binding to CBP. Also acts as a putative regulator of mRNA stability and/or translation rates and mediates mRNA nuclear export.

Descripción

KHDRBS1, also known as Sam68 (Src-associated in mitosis, 68 kDa), is an RNA-binding protein that contains a KH (hnRNP K homology) domain, enabling it to regulate RNA processing, splicing, and translation. It interacts with signaling proteins such as Src and is involved in linking signal transduction pathways to RNA metabolism. KHDRBS1 plays roles in cell cycle progression, proliferation, and apoptosis, particularly during mitosis, where it is phosphorylated by Src kinases. It is expressed in multiple tissues, particularly in proliferating cells and neurons, and regulates mRNA stability and alternative splicing of key genes like BCL2 and CD44. Dysregulation of KHDRBS1 is associated with cancers, including breast and prostate cancer, where it promotes tumor progression and metastasis by modulating RNA processing of oncogenic transcripts. Knockdown studies reveal defects in cell division, proliferation, and stress responses, highlighting its dual role in signal transduction and RNA regulation.
