ACOT12 is a cytosolic enzyme that hydrolyzes short- and medium-chain acyl-CoA esters, contributing to lipid metabolism and detoxification pathways. It is expressed in tissues such as the liver, kidney, and adipose tissue, where it regulates intracellular fatty acid levels and prevents the accumulation of toxic acyl-CoA intermediates. ACOT12 plays an essential role in modulating lipid signaling molecules and providing precursors for biosynthetic pathways, including membrane lipid synthesis. Additionally, ACOT12 influences metabolic flexibility by ensuring the proper balance between fatty acid oxidation and lipid storage during fed and fasting states. Dysregulation of ACOT12 has been linked to metabolic disorders, including non-alcoholic fatty liver disease and obesity, where altered lipid metabolism affects systemic energy homeostasis.
