Descripción
HTR1D is a member of the serotonin receptor family and is classified as a G protein-coupled receptor (GPCR) that mediates the inhibitory effects of serotonin (5-HT). Predominantly located in the central nervous system, particularly in regions like the trigeminal nerve and cerebral vasculature, HTR1D plays a crucial role in regulating vasoconstriction, pain modulation, and neurotransmitter release. It functions primarily as an autoreceptor, inhibiting serotonin release to maintain neurotransmitter balance. HTR1D has been strongly implicated in the pathophysiology of migraine headaches, where it mediates the constriction of intracranial blood vessels, reducing pain transmission. Triptan drugs, such as sumatriptan and zolmitriptan, act as selective HTR1D agonists and are widely used for the acute treatment of migraines by targeting this receptor to alleviate vasodilation and inflammation. Beyond migraines, HTR1D is also involved in modulating anxiety, mood, and nociception (pain perception). Dysregulation of this receptor can contribute to neuropsychiatric disorders such as depression and anxiety disorders. The study of HTR1D as a target for novel therapeutics offers opportunities for better migraine management and potential treatments for mood disorders through modulation of serotonin pathways.
