Descripción

UBASH3B, also known as STS-1 (Suppressor of T-cell receptor signaling 1), is a negative regulator of T-cell receptor (TCR) signaling that functions by dephosphorylating signaling intermediates and attenuating downstream activation of NF-κB and MAPK pathways. It contains ubiquitin-associated (UBA) and SH3 domains that mediate interactions with signaling proteins and ubiquitinated molecules, promoting the regulation of protein stability and turnover. UBASH3B is expressed predominantly in immune cells, where it plays a role in T-cell activation, immune tolerance, and inflammation control. Dysregulation of UBASH3B is associated with autoimmune diseases and impaired immune responses due to excessive TCR signaling. Knockout studies reveal hyperactive T-cell signaling, increased cytokine production, and heightened immune activation, underscoring its role in maintaining immune homeostasis by regulating TCR signaling and protein ubiquitination pathways.
