Diphthamide biosynthesis 3 pseudogene 1 (DPH3P1) is categorized as a pseudogene, derived from the functional DPH3 gene involved in diphthamide biosynthesis, a unique post-translational modification of histidine residues in elongation factor 2 (EF2). While the parental DPH3 gene actively participates in the enzymatic pathway, the pseudogene DPH3P1 does not code for a functional protein, but its sequences might still have regulatory roles at the transcriptional or post-transcriptional level. Pseudogenes like DPH3P1 can influence the expression of their parent genes by acting as competitive endogenous RNAs (ceRNAs), sequestering microRNAs, or through epigenetic modifications, although direct evidence in this case is yet to be fully substantiated. DPH3P1 is an intriguing element in genomic research, shedding light on pseudogene functionality beyond their initial categorization as "non-functional" remnants. Understanding its role could reveal new dimensions in gene regulation and potential implications in diseases where diphthamide biosynthesis pathways are disrupted.
