P2RY12 is a G protein-coupled receptor activated by ADP, playing a crucial role in platelet aggregation and thrombus formation. P2RY12 couples to Gi proteins, inhibiting adenylate cyclase, reducing cAMP levels, and enhancing platelet activation and aggregation in response to ADP. It is highly expressed in platelets and some immune cells, where it regulates hemostasis, inflammation, and thrombosis. P2RY12 is a major target for antiplatelet drugs such as clopidogrel, prasugrel, and ticagrelor, which are used to prevent arterial thrombosis, stroke, and myocardial infarction by inhibiting platelet activation. Dysregulation of P2RY12 contributes to thrombotic disorders, while excessive inhibition can lead to bleeding complications. Beyond its role in hemostasis, P2RY12 is involved in immune cell chemotaxis and inflammatory responses. The therapeutic targeting of P2RY12 continues to evolve, particularly for the management of cardiovascular diseases, thrombosis, and inflammation.
