Descripción
KCTD8 is a member of the potassium channel tetramerization domain (KCTD) family, known for its role as an auxiliary subunit of GABA-B receptors. It modulates the pharmacology and kinetics of GABA-B receptor signaling, influencing inhibitory neurotransmission in the central nervous system. KCTD8 binds to the GABA-B receptor complex, regulating receptor activation, desensitization, and downstream signaling pathways mediated by Gi/o proteins. It is primarily expressed in brain regions involved in cognition, motor control, and emotional regulation, contributing to synaptic plasticity and neuronal excitability. KCTD8 has also been implicated in mood disorders, such as anxiety and depression, where dysregulation of GABA-B signaling disrupts inhibitory balance. Beyond neurotransmission, KCTD8 participates in protein-protein interactions through its BTB/POZ domain, regulating cellular processes like ubiquitination and transcriptional repression. Altered expression of KCTD8 has been associated with neurological disorders and certain cancers. Its function as a GABA-B receptor modulator positions KCTD8 as a target of interest for improving treatments for neurological and psychiatric conditions.
