Background

KCTD8 is a member of the potassium channel tetramerization domain (KCTD) family, known for its role as an auxiliary subunit of GABA-B receptors. It modulates the pharmacology and kinetics of GABA-B receptor signaling, influencing inhibitory neurotransmission in the central nervous system. KCTD8 binds to the GABA-B receptor complex, regulating receptor activation, desensitization, and downstream signaling pathways mediated by Gi/o proteins. It is primarily expressed in brain regions involved in cognition, motor control, and emotional regulation, contributing to synaptic plasticity and neuronal excitability.
