DNA fragmentation factor subunit beta (DFFB) is an essential component of the apoptotic machinery, functioning as a part of the DNA fragmentation factor (DFF) complex. DFFB forms a heterodimer with its partner subunit, DNA fragmentation factor subunit alpha (DFFA). This beta subunit possesses the enzymatic activity required for the cleavage of DNA into nucleosomal fragments, a hallmark of apoptosis. DFFB, a caspase-activated DNase, is kept in an inactive state by its binding to DFFA until caspases, triggered by apoptotic signals, cleave DFFA, releasing DFFB to execute its DNase function. Dysregulation of DFFB activity is associated with pathological conditions, including resistance to apoptosis in cancer and excessive cell death in degenerative diseases. DFFB activity is tightly regulated to ensure its apoptotic function is confined to specific cellular contexts.

The apoptotic process is accompanied by shrinkage and fragmentation of the cells and nuclei and degradation of the chromosomal DNA into nucleosomal units. DNA fragmentation factor (DFF) is a heterodimeric protein of 40-kD (DFFB) and 45-kD (DFFA) subunits. DFFA is the substrate for caspase-3 and triggers DNA fragmentation during apoptosis. DFF becomes activated when DFFA is cleaved by caspase-3. The cleaved fragments of DFFA dissociate from DFFB, the active component of DFF. DFFB has been found to trigger both DNA fragmentation and chromatin condensation during apoptosis.
