Background

CXCR5 is a G protein-coupled receptor expressed mainly on follicular B cells and T follicular helper (Tfh) cells, where it binds its ligand CXCL13. CXCR5 is critical for B cell homing to lymphoid follicles within secondary lymphoid organs, such as lymph nodes, spleen, and Peyer’s patches. It guides the migration of B cells and Tfh cells to germinal centers, facilitating antibody production, affinity maturation, and immune memory formation. CXCR5 signaling activates pathways like PI3K and ERK, driving cell migration and positioning within lymphoid tissues. In autoimmune diseases, such as systemic lupus erythematosus (SLE) and rheumatoid arthritis, dysregulated CXCR5 expression contributes to abnormal germinal center formation and autoantibody production. CXCR5 is also involved in chronic inflammatory diseases where it promotes immune cell accumulation. In cancer, CXCR5 expression is observed in B cell lymphomas and some solid tumors, where it enhances tumor growth, immune evasion, and metastasis. CXCL13-CXCR5 signaling in the tumor microenvironment supports lymphoid tissue-like structures that promote tumor progression. Therapeutically, targeting CXCR5 is being explored to modulate B cell activity in autoimmune diseases and lymphomas. CXCR5 is essential for coordinating B and Tfh cell migration, ensuring proper immune responses while contributing to disease pathology when dysregulated.
