Nuclear receptor subfamily 1 group H member 4 (NR1H4), also known as FXR (farnesoid X receptor), is a nuclear receptor that plays a crucial role in regulating bile acid metabolism, liver function, and glucose homeostasis NR1H4 is expressed primarily in the liver, intestine, and kidneys, where it regulates the synthesis, transport, and catabolism of bile acids, which are essential for digestion and the absorption of fats FXR also influences lipid and glucose metabolism, helping maintain energy balance and insulin sensitivity in the liver and other metabolic tissues It plays a key role in protecting the liver from bile acid toxicity by regulating the expression of genes involved in bile acid homeostasis Dysregulation of FXR signaling has been associated with liver diseases such as non-alcoholic fatty liver disease (NAFLD), cirrhosis, and cholestasis, as well as metabolic diseases like obesity and diabetes FXR has emerged as a therapeutic target for managing liver diseases, metabolic syndrome, and conditions related to bile acid imbalances, making it an important receptor for drug development in these areas
