Glucosaminyl (N-acetyl) transferase 4 (GCNT4) is a glycosyltransferase enzyme responsible for catalyzing the formation of core 2 O-glycans, critical components of glycoproteins and mucins involved in cellular adhesion, signaling, and epithelial protection GCNT4 is expressed primarily in the gastrointestinal tract, pancreas, and other mucin-secreting tissues where it contributes to the maintenance of epithelial integrity and mucus production Studies have shown that GCNT4 plays a role in regulating glycoprotein branching, which is essential for cell surface interactions and immune responses Dysregulation of GCNT4 has been linked to colorectal and pancreatic cancers, where altered glycosylation promotes tumor growth, metastasis, and resistance to apoptosis Furthermore, GCNT4 expression is modulated during inflammatory conditions, impacting mucosal defense and contributing to diseases like ulcerative colitis and Crohn’s disease Emerging evidence highlights its role in epithelial homeostasis and its potential as a biomarker for gastrointestinal diseases and therapeutic target for cancer treatment
