Descripción

BLNK (B-cell linker) is an adaptor protein primarily expressed in B cells, where it plays a crucial role in regulating B cell receptor (BCR) signaling. It functions by linking various signaling molecules, which is critical for the proper activation, differentiation, and survival of B cells. Upon BCR engagement, BLNK is phosphorylated, leading to the recruitment of other signaling proteins, including those involved in the activation of key pathways such as the phosphoinositide 3-kinase (PI3K) and mitogen-activated protein kinase (MAPK) pathways. This allows BLNK to help orchestrate B cell activation and the immune response. It is also involved in regulating B cell proliferation and the transition from the naive to the activated state, essential for effective immune responses. Mutations in BLNK can result in immune dysfunction and are associated with immune-related diseases, including lymphoid malignancies. Understanding its role in B cell signaling can help inform therapies for diseases involving B cells, such as autoimmune diseases and B-cell malignancies.

Plays a role in the activation of ERK/EPHB2, MAP kinase p38 and JNK. Modulates AP1 activation. Important for the activation of NF-kappa-B and NFAT. Plays an important role in BCR-mediated PLCG1 and PLCG2 activation and Ca(2+) mobilization and is required for trafficking of the BCR to late endosomes. However, does not seem to be required for pre-BCR-mediated activation of MAP kinase and phosphatidyl-inositol 3(PI3) kinase signaling. May be required for the RAC1-JNK pathway. Plays a critical role in orchestrating the pro-B cell to pre-B cell transition. May play an important role in BCR-induced B-cell apoptosis.
